He claimed 27 match victories to finish 11 points ahead of second-place Humphries at the top of the standings, with Nathan Aspinall and Gerwyn Price also joining them in the top four.
Tonight's action in London will get underway at 7.15pm, with Littler taking on Price in the first match as the players go head-to-head in a best of 19 encounter. Humphries and Aspinall will then battle it out in the second semi-final.
The showpiece match later in the evening will be played as a best of 21 encounter, with the winner set to pocket a whopping £275,000 in prize money.
"It's been a good 16-week campaign but that is all out the window now. I finished first once again and now I just can't wait," Littler said ahead of the Play-Offs.
"Obviously the stats are there to be shown. I've dominated in many ways, the averages, 180s, ton-plus checkouts - but it's down to one night."
